Container / Specimen Requirements

    Container / Tube: Yellow serum separator Vacutainer® tube
or Green Vacutainer® tube (lithium heparin) or
Red Vacutainer® tube.
    Minimum Volumes: 1 ml
Special Handling or Timing of Specimen:

1 Hour GTT

Blood is collected 60 minutes after the subject completes drinking the glucose tolerance beverage.

2 Hour GTT

Blood is collected as fasting, 0.5, 1, 1.5, and 2 hour specimens.

3 Hour GTT

Blood is collected as fasting, 1, 2, and 3 hour specimens.
Preparation of Patient:

Patient must be fasting for at least 10 hours but not more than 12 hours preceding the test. Patient should avoid coffee, smoking and unusual physical activity for at least 8 hours prior to the test. Patient should not smoke during the test. Water is allowed.

Patient must drink the glucose tolerance beverage within 5 minutes.

Patients are dosed according to the following rules:  

1 Hour GTT

150 mL Dextrol beverage. 1/2 bottle = 50 grams glucose.

2 Hour GTT

All non-pregnant patients: 225 mL Dextrol beverage. ¾ bottle = 75 grams glucose

3 Hour GTT

Pregnant patients: 300 mL Dextrol beverage. One full bottle = 100 grams glucose.

Non-pregnant patients: 225 mL Dextrol beverage. ¾ bottle = 75 grams glucose.

Reference Range

 

1 Hour GTT

 

For a 50-gram oral glucose tolerance test (150mL Dextrol) used to screen for gestational diabetes, normal blood values at 1 hour are less than 140 mg/dL.

2 Hour GTT

 

For a 75-gram oral glucose tolerance test (225mL Dextrol) used to check for type 2 diabetes, normal (non-diabetic) blood values are:

· Fasting: 60 to 110 mg/dL (adults)
· Fasting: 60 to 130 mg/dL (children)
· ½ hour: less than 200 mg/dL
· 1 hour: less than 200 mg/dL
· 1 1/2 hour: less than 200 mg/dL
· 2 hours: less than 140 mg/dL.

Note: Children less than 16 years of age are dosed according to their weight.

 

3 Hour GTT

 

For a 100-gram oral glucose tolerance test (300 mL Dextrol) used to screen for gestational diabetes, normal blood values are:
· Fasting: less than 95 mg/dL
· 1 hour: less than 180 mg/dL
· 2 hour: less than 155 mg/dL
· 3 hour: less than 140 mg/dL

For a 75 gram oral glucose tolerance test (225mL Dextrol) used to check for diabetes.
· Fasting: 60 to 110 mg/dL
· 1 hour: less than 200 mg/dL
· 2 hours: less than 140 mg/dL.
· 3 hours: less than 140 mg/dL

 

Causes for Specimen Rejection:  Improper labeling, patient not fasting, and/or hemolysis.
